On a Canon 80D, I found an option called “IS start” under Custom Controls for the depth-of-field preview button. What does “IS” stand for, and what happens if I assign this function to a button?

The image is of a shaking hand — I and S stand for "image stabilization".

This is explained on page 439 of the manual (in English):

IS start

If you press the button assigned to this function when the lens’s IS switch is set to <ON>, the lens’s Image Stabilizer will operate.

If you don't know what the initials stand for, or can't guess what the icon might mean, a search for the phrase "IS Start" in in the manual found on Canon's support page for the 80D will jump right to it.

“IS” stands for Image Stabilization. On Canon, assigning “IS start” to a button means that pressing that button will activate the lens’s image stabilizer, as long as the lens itself has IS and its IS switch is set to ON.

Normally, Canon lenses activate IS when metering is active, often with a half-press of the shutter button or when using back-button focus. The camera then stops IS after metering times out to save battery power.

So “IS start” is simply a way to start stabilization from a specific custom button instead of relying only on the usual shutter-half-press behavior.
