My Canon EOS 450D appears to autofocus incorrectly when using the optical viewfinder, but focus is accurate in Live View (though slower). The issue became obvious after I bought a Canon 50mm f/1.8 II: through the viewfinder it seems to front-focus badly, even at longer distances.

I’ve heard there may be an adjustment screw behind the mirror, but I’m worried about damaging the camera. I also don’t have a nearby service center. Is there any safe user adjustment or software method to correct this, or does the camera/lens need professional calibration?

I would suggest to take it to a service center for calibration. If you do it yourself you run the risk of totally mis aligning it. It could also be a lens issue since you say that it only happens with that lens.
