While shooting, my Canon 1D X worked normally for about 30 frames, then on one exposure the shutter stayed open for a couple of seconds and the camera displayed Err 01, indicating a communication problem with the lens. After half-pressing the shutter, the message cleared and the camera worked normally for the rest of the day.

Back home I removed the lens, checked it, and used a blower around the mount/contacts. I haven’t seen the error again.

If this only happened once, is it likely just a temporary lens/body contact issue, or is it a sign that the camera or lens should be serviced? At what point would repeated Err 01 messages justify sending the body or lens in for inspection?

It's a lens to body communication error and may be as simple as some dirty contacts. If you clean these and the error comes back and persists then your lens and/or body may need servicing. Otherwise, I think you probably don't need to worry, especially since it has been fine ever since.
