I’m trying to use a Godox 300Di studio strobe with a Canon EOS R5 Mark II and I’m unsure which flash trigger will work.
Can this light be triggered directly by a Godox radio transmitter, or do I need a separate receiver connected to the strobe? If a receiver is required, what type of sync connection does the 300Di use?

The 300Di is one of the "Mini Pioneer" series of Godox strobes, which does not have built-in radio remote control, so there is no Godox radio transmitter that could trigger the strobe by itself. But there are several ways you can fire it in sync with your shutter.
Cable a receiver to the strobe
The strobe needs to be cabled to a receiver the radio transmitter can "speak" to. Whatever triggers you use, though, if you cable them to the strobe can only tell the strobe to fire. There will be no other remote control over the flash.
The sync port on the 300Di is 3.5mm, below the on/off switch. So you need to find a cable that matches whatever receiver you choose on one end and 3.5mm on the other. For example, if you choose a Godox X1R receiver, it uses 2.5mm, so you'd need a 2.5mm to 3.5mm cable to connect it to the 300 Di. And then you could fire the 300Di over radio with any Godox "X"-named transmitter on the hotshoe of your camera.
Trigger the strobe by its optical sensor with another flash
The 300Di also has a light sensor (the little red thing sticking up on the back end) that can trigger the light if another flash burst goes off (say, from a pop-up/built-in flash on your camera or another flash on your camera hotshoe). This "master" flash must be in M without any preflahes, and the SLAVE mode must be turned on on the 300Di.
Cable the strobe to your camera
You can also use a cable. Your R5 Mark II has a 2.5mm flash sync port below its 3.5mm headphone port. So, you could also get a long 2.5mm to 3.5mm mono audio cable to connect your camera directly to the 300DI to fire it.
Additional notes about using a Mini Pioneer or E-series Godox strobe
The 300Di does not have a modifier mount to attach a softbox to. If you wish to use a Bowens-mount modifier (e.g., softbox, octa), you will need to find one of these clamp-on adapters to get a Bowens mount to bayonet-mount a modifier to.
Today, in late 2025, I tend to recommend a Godox MS300V line strobe instead of a Mini Pioneer or E-series as a 300 Ws low-cost manual AC Godox monolight (US$130 new), because it has built-in radio remote control in the Godox 2.4 GHz system (not just firing, but also M power adjustment by group and modeling light control) and a Bowens S mount on the face. It's much newer and easier to use.
