I want to use a Nikon EL-Nikkor 50mm f/2.8 enlarging lens on a Canon 5D Mark II. Since it is an enlarger lens rather than a standard camera lens, what kind of adapter or reversing setup is needed to attach it and use it on the camera?

The lens in question is designed for a photographic enlarger which is used during film printing to enlarge the negative onto the photo paper. However, there are places that will, in fact, make adapters one of which is Zörk Film and Phototechnic in Germany. They don't really list the product detail, but if you contact them, they can probably let you know if it is possible. I believe the Multi-Focus System uses enlarger lenses.
For interest, Nikon talks about this lens here: http://www.nikon.com/about/feelnikon/recollections/r15_e/index.htm

The EL-Nikkor 50mm f/2.8 is an enlarger lens, so it does not mount directly to an EF camera body like the 5D Mark II. In practice, people typically use it with a custom or improvised mount plus extension tubes, often for close-up or macro work.
Based on the answers, workable options include:
- Mounting the lens to a modified Canon body cap, then attaching that to Canon extension tubes.
- Using a Canon-to-M42/Pentax screw adapter on the camera, then M42/Pentax extension tubes, and mounting the enlarger lens in reverse with a custom adapter or modified body cap.
- Looking for a specialist custom adapter maker such as Zörk.
Important points:
- This is usually a DIY/custom-adapter setup, not a simple off-the-shelf EF mount solution.
- The aperture ring can be stiff, so the lens must be secured firmly so it does not twist loose.
- Because enlarger lenses are meant for enlargers, expect manual operation only and likely use with extension tubes rather than normal infinity-focus photography.
